OPK and DPO

We used an OPK this month. Do you count the day after your pos OPK as your DPO or do you wait a day since it can take 24 hrs to ovulate. Ex: Pos OPK on thur. Is Fri my first DPO or if Sat? Does that make any sense? Does it really matter?
